If you are talking about romantic relationships I see Giles/Modesty or Willie/Faith as being most likely. With Xander, Modesty, Willie or Buffy I see friendship as the most likely theme. A Xander/Willie or a Buffy/Modesty buddy story could be fun. Maybe with Andrew or Dawn along as well for friction.
